I found rodent damage to wiring in my truck and several systems not functioning properly. I suspect my comprehensive coverage will help but do I reach out to my agent before or after taking it to a shop/dealership for an estimate?

You talk to the claims department. Your agent isn’t s claims adjuster. Don’t be disappointed if they say they don’t cover it. I’d be surprised if they do.
My mom’s car wiring was damaged by rats. Covered under her comprehensive policy. But you could get an estimate and decide if you want to pay or if pocket or file a claim. Apparently this is not uncommon.
